In this study, silk fibroin and Beta Cyclodextrin composite scaffolds will be fabricated using a freeze-drying method for effective wound dressing and drug-releasing application. The prepared Beta Cyclodextrin and silk fibroin 3D scaffold will be loaded with ciprofloxacin HCL which is a soluble antimicrobial drug and commonly used in tissue engineering. Beta Cyclodextrin, which also acts as a drug carrier may carry and release Ciprofloxacin HCL to the wound site for a certainly long duration of time. The prepared composite scaffolds will be characterized by Fourier Transform Infrared (FTIR) spectroscopy, Scanning Electron Microscopy (SEM), X-Ray Diffraction (XRD) and evaluated for antibacterial properties. These composite scaffolds using silk fibroin, Beta Cyclodextrin with an appropriate antimicrobial agent could successfully inhibit the growth of bacteria. Therefore, Green composite could be a promising candidate for wound healing applications.

Project Objectives- Preparation of Cyclodextrin incorporated and Ciprofloxacin loaded antimicrobial composite for wound healing.
- To evaluate antibacterial activity by Agar Diffusion test method.
- Structural and surface characterization of prepared scaffolds using SEM, FTIR, and EDX method
- Characterization and Comparison of Cyclodextrin integrate double and chitosan-based single carrier composite for wound healing.
This project is divided into three stages. The first stage is to extract silk fibroin and eliminate sericin protein from Bombyx mori silk cocoons by degumming process and obtain silk fibroin solution by chemical treatment. The second stage requires the silk fibroin solution to be mixed with Beta Cyclodextrin using citric acid as a cross-linking agent to prepare a uniform mixture of Beta Cyclodextrin and silk fibroin. This mixture would be transferred to Petri dishes and frozen overnight. At last, the freeze-dried plates would be lyophilized for 24 hours and 3D composite scaffolds are obtained. The prepared silk fibroin composite scaffold would then be loaded with 0.5% Ciprofloxacin solution and dried at room temperature. Lastly, the yield composite product would be characterized using FTIR, SEM, and XRD and tested for its antibacterial activity.
Benefits of the ProjectIn recent research studies, there is one of the most serious challenges to prevent the infection by controlling drug delivery system. In the perspective of the clinical applications mostly the antibacterial drugs, antibiotics are given to the patients for infection prevention which is fatal for the patient health because the antibiotics and oral drugs have too much side effect as compared to the dressing drugs, but also dressing drugs facing problem in drug-releasing properties which is important for infection healing. However, this project provides a controlled and long durable antibiotic drug release system on the 3D composite scaffold to minimize infections in chronic wounds due to Beta Cyclodextrin. These composite scaffolds are expected to provide a slow antibiotic release to the wound site leading to rapid wound healing. Thus, these scaffolds will prevent the body from harmful side effects of antibiotics and low quality dressing drugs also solve the problem of the releasing drug for a certain duration of the time.
Technical Details of Final DeliverableThe project comprises the preparation of silk fibroin and Beta Cyclodextrin composite scaffolds. The prepared composite scaffolds for wound dressing will include surface and structural characterization using Fourier Transform Infrared (FTIR) spectroscopy, Scanning Electron Microscopy (SEM), and X-Ray Diffraction (XRD).
